All COCs from each dog were randomly divided into fi ve groups. Every group except the control (group 1) was equilibrated and separately introduced into a different recipe for the vitrifi cation medium. A vitrifi cation medium consisting of tissue culture medium (TCM) 199, supplemented with 0.5 M trehalose, 1.5 M ethylene glycol and 20% fetal bovine serum was used for COCs for groups 2 and 4, whereas a vitrifi cation medium consisting of TCM 199, supplemented with 0.5 M trehalose, 3 M ethylene glycol and 20% fetal bovine serum was used for groups 3 and 5. Only the COCs from groups 4 and 5 were vitrifi ed with the solid surface method. The survival rates of non-vitrifi ed and vitrifi ed COCs were not signifi cantly different. Morphological alterations including shrinkage, disrupted oolemma, cumulus cell damage and cytoplasmic vacuolization were common features found in treatment groups. The percentages of COCs with each type of damage were not different. However, cumulus cells appeared to be the structure most affected by the SSV method. |

At follicular phase, POEC contained a great number of long ciliated cells whereas those at luteal phase consisted mostly of round shaped nonciliated cells with short microvilli on the apical surface. This change in morphological features of POEC seemed to serve well on their functions as oocyte transporters at follicular phase. As for COCs, they were morphologically classified into 4 types based on the accumulation and arrangement of cumulus cells around the oocytes. These were intact cumulus cell layer, single cumulus cell layer, partial cumulus cell layer and completely denuded oocytes at the percentages of composition of 19.87%, 18.13 %, 28.88 % and 33.12 %, respectively. Cumulus cells attached on the surface of oocytes were teardrop-like shape having the conical ends pointed towards the oocytes membrane surface while free-floating cumulus cells in the follicular fluid and those at the outer layers were round in shape. These POEC and COCs were high potential feeder cells and could be further cultured for in vitro fertilization use. |

Each group was received exogenous hormone, the human chorionic gonadotropin at 0, 1, 3, 6, 9 and 12 I.U./gm.. After thirty day of experiment, those who received the hormone at 1, 3 and 6 I.U./gm. showed no sign of ovarian development. Their ovaries still maintained at stage I, and consisted mainly of oogonia and immature oocytes when observed under light microscope. This results was similar to those of sham control. However, ovaries of those who received the hormone at 9 and 12 I.U./gm. developed into stage II and stage III and consisted mainly of early mature oocyte and mature oocyte. |
