A series of staining procedures were employed including peroxidase - labeled lectin - diaminobenzidine methods and digestion techniques with neuraminidase or ? - amylase. The terminal portion of the anterior lingual gland consisted of mucous and seromucous cells. All the mucous cells in the glands possessed a mixture of glycoconjugates of different nature, but they could be divided into three types, on the basis of the principle type of glycoconjugates involved, namely primarily sulfated, carboxylated and neutral glycoconjugate-containing cells. The first mentioned type of cells was predominant. The presence of sialic acid, ? - D - mannose, ? - D - glucose and ? - D - galactose in these glycoconjugates was confirmed. The seromucous cells found in the anterior lingual gland contained a very small amount of comparable carbohydrates predominantly in the apical cytoplasm. Possible functional activities of the glands were discussed. |

The staining procedures employed were alcian blue (AB) pH 1.0 AB pH 2.5, Periodic acid Schiff (PAS) and AB pH 2.5-PAS. In addition twelve lectins, Triticum vulgaris WGA, Concana valin A, Glycine max SBA, Dolichos biflorus DBA, Arachis hypogea PNA, Solanum tuberosum STL, Datura stramonium DSA, Ulex europeus UEA-I, Bauhinia purpurea BPA, Ricinus communis RCAI, Lotus tetragonolobus LTA, and Limulus polyphemus LPA were applied to detect saccharides residues. According to the results obtained, the nature of glycoconjugates in mucous epithelium was found to change during cell migration from deep to superficial part. The surface mucous cells contained primarily neutral glycoconjugates with vicinal diol groupings and terminal fucose residues. By contrast, the mucous cells in deep pits and in pyloric glands contained acidic sulfated glycoconjugates with terminal sialic acid residues. The ?-D-mannose, ?-D-glucose, ?-D-galactose, N-acetylglucosamine and N-acetylgalactosamine were also found at the pyloric gland cells while they were smaller in amount at the surface mucous cells. The data suggested that changes of epithelial cells were closely correlated with cellular maturation. |

The lectins used in the present study were Concanavalin A (Con A), Ricinus communis agglutinin-I (RCA-I), wheat germ agglutinin (WGA), Dolichos biflorus agglutinin (DBA), Ulex europeues agglutinin - I (UEA-I), Limax flavus agglutinin (LFA), Lotus tetragonolobus agglutinin (LTA) and peanut agglutinin (PNA). According to the results obtained, acidic and neutral glycoconjugates with ?-D-mannose, ?- D-glucase, ?-D-galactose, N-acetyl-D-glucasamine, ?-L-fucose and sialic acid residues were visualized in the secretory cells of chicken mandibular gland. |

The results showed both mucous and serous cells in secretory endpieces of sublingual salivary gland. Mucous cells showed a large number of vicinal diol groups with acid and neutral glycoconjugates. Lectin histochemistry gave evidence of the presence of mannose, ? N - acetylgalactosamine, N- acetylglucosamine and sialic acid linked to Galactosyl (?13) N – acetylgalactosamine residues. Serous cells had positive reaction with almost all the conventional employed, with the exception of acid glycoconjugates which were less diffused. Serous cells secreted predominantly neutral glycoconjugates with mannose, Galactosyl (?14 ) N- acetylglucosamine, N- acetylglucosamine and fucose residues. |

The quantity of neutral glycoconjugates with ?-D-mannose, ?-D-glucose, ?-D-galactose and N-acetylglucosamine increased from mucosal crypt up to mucosal surface of the epithelium, while sialic acid residues and sulfated glycoconjugates decreased from mucosal crypt to mucosal surface. |

The lectin used in the present study were peanut agglutinin (PNA), wheat germ agglutinin (WGA), Dolichos biflorus agglutinin (DBA), and Lotus tetragonolobus agglutinin (LTA) The mucosa of both coprodeum and urodeum is covered by simple columnar epithelium in which 2 type of cells are encountered-columnar and goblet cells. In contrast, one layer of only columna cells is found in the mucosa of proctodeum. Tubular glands of proctodeum are commonly provided at submucosal layer. All the goblet cells in coprodeum and urodeum and tubular glands of proctodeum were coloured blue and purple with AB pH 1.0 and AB pH 2.5-PAS, respectively. They were furthermore stained moderately with WGA. Neuraminidase digestion changed the blue coloration with AB pH 2.5 to pale blue. Removal of sialic acid with neuraminidase imparted weak to strong affinity of PNA. Histophysiological significances of glycoconjugates involved in the chicken cloaca were discussed with special reference to the functional activities of the carbohydrates. |
