The effect of pectinase enzyme concentrations (0-0.2%) and incubation times (60-300 min) on juice yield (%), total soluble solids (TSS), recovery soluble solids (RSS), clarity (%T670) and browning index (A420) of juice were studied using central composite design of experiments. Results showed that pectinase enzyme concentration played an important role that significantly (p ? 0.001) influenced most of dependent variables of banana juice. The coefficient of determination (R2) of yield (%), recovery soluble solids (RSS), clarity (%T670) and browning index (A420) were 0.907, 0.924, 0.804 and 0.793, respectively. The optimum condition for enzymatic extraction was 0.15% of pectinase enzyme incubated for 120 min at 50?C. The yield was ? 62%, RSS ?14, %T670 ? 96% and A420 ? 0.1. The physicochemical compositions of syrup were mostly three times higher than juice owing to the concentration that increased from 24 to 74? Brix except for pH, clarity (%T670), CIE L* which were not changed. |

Strain selection was performed by using solid substrate fermentation composing of wheat bran and rice husk in a ratio of 18:2. It revealed that Rhizopus sp. 26R gave almost the highest activity in 2 days and remained constant for 4 days. Optimization studies showed that addition of either 1 g raw cassava starch or pectin to a 20 g substrate increased the enzyme activity to 1.7 and 2.4 times, respectively. The highest activity, however, obtained when the strain was grown on substrate containing a mixture of wheat bran, rice bran and rice husk in a ratio of 9:9:2 or 6:12:2 with 58% initial moisture content, pH adjusted to 5.7 and incubation was at 32 degree C. Under these conditions, Rhizopus sp. 26R produced ca. 700 units of enzyme activity per gram of wet weight substrate. |

The fermentation quality of fermented cocoa beans with added pectinase were shown to be more promising than those without enzyme batch . The amount of full fermented beans (4/4F) were increased concomitantly with the increasing of enzyme soaking period. Full fermented beans were found at 79 and 86% of fermented beans in enzyme – soaking batches of 3 and 6 hours respectively. No slaty and mold beans were observed in these batches. This indicated that the quality of fermented beans was equivalent to standard grade 1. Additionally, it was found that the pH levels of full fermented beans were 5.49 and 5.92 which were acceptable in the cocoa market. Pectinase –producing microorganism therefore, would have an important role in cocoa fermentation processes. |
