Abstract

Lipid peroxidation (LPO) is the process whereby oxygen free radicals steal electrons from polyunsaturated fatty acids of cell membranes, initiating a self-propagating chain reaction and the end-products of the reactions are dangerous to the viability of cells. It is used as an indicator of oxidative stress or the amount of free radicals in cells and tissues. The observation of LPO activities in various organs of mud crab (Scylla serrata) over the molting cycle showed evidence that supported the functions of free radicals. LPO in the intermolt stage was significantly (P < 0.05) lower than at the premolt stage. The LPO activities gradually increased when the crab entered the premolt stage which was detected in the haemolymph, gill, hepatopancreas, integument and muscle of crabs. The hepatopancreas seemed to be the organ where the free radical processes occurred. LPO in the haemolymph, integument and gills had the highest levels 2 d before molting while the activities in the hepatopancreas showed distinguishing functions 2 wk before molting. The determination of antioxidant activity using free radical scavengers showed the same pattern in the gills, meat, hepatopancreas and claws. The activities gradually decreased from the intermolt stage until they reached a minimum in the early postmolt, and then increased again at late postmolt. It clearly indicated that LPO activities in the molting cycle corresponded with the physiology during the stage changing and adaptation of crab.

Abstract

The activities of the antioxidative enzyme, catalase (CAT), and the non-enzyme, glutathione (GSH) in 12 stages over the molting of mud crab (Scylla serrata) collected from Chanthaburi Province were determined. The stages were defined as: A1 (6-hour postmolt), A2.1 (12-hour postmolt), A2.2 (24-hour postmolt), B1 (2-day postmolt), B 2.1 (3-day postmolt), B2.2 ( 5-day postmolt), B2.3 (7-day postmolt), B2.4 (10 day postmolt), C (intermolt), D1 (2-week premolt), D2 (1- week premolt) and D3 (2-day premolt). The results revealed that CAT which controlled and eliminated excessive H2O2, increased in premolt (D1–D3) then dropped in the early postmolt stages (A1–A2.2) and increased again in the late postmolt stages (B1–B2.4). GSH, a reducing substance, which is also involved in the detoxification of H2O2 showed the same pattern in most tissues except in the hepatopancreas where it increased from the early postmolt stage (A1) and continued to increase through the postmolt stages. The variation in activity of CAT and GSH corresponded well with the metabolic activity through the molt cycle.

Abstract

Activity profiles of chitobiase, proteinase and the content of glycogen and trace elements in gill, integument and haemolymph of mud crab (Scylla serrata) during molting cycle were determined. Chitobiase activity was at the highest level of 0.5467?0.0136 ?mol min-1mg protein-1 at 1-week premolt (D2) in gill and at 0.1413?0.0026 ?mol min-1 mg protein-1 in integument but reached the maximum level of 0.0851?0.0123 ?mol min-1mg protein-1 at 24-h postmolt (A2.2) in haemolymph. The high activities of proteinase, on the other hand, were found to be 0.6511 ? 0.0934 units mg protein-1 at 7-day postmolt (B2.3), 0.1553 ? 0.0419 units mg protein-1 at 1-week premolt (D2), and 0.1827?0.0110 units mg protein-1 at 12-h postmolt (A2.1) in gill, integument and haemolymph, respectively. These two enzymes are needed for carapace degradation as well as for chitin synthesis in preparation for carapace formation. For glycogen content, high accumulation of glycogen in the integument was observed throughout the molting cycle except at intermolt (C). In gill, however, the level of glycogen was high only before molting began (D2) while glycogen content in hepatopancreas and muscle, energy reserve tissues, was rather fluctuated but reached the maximum levels at 24-h postmolt (A2.2). As for trace elements, copper, a major component of hemocyanin, was found at high level at intermolt stage (C) in all tissues while magnesium, involving both nerve conduction and muscle contraction, showed the highest content at 7-day postmolt (B2.3) and 1-week premolt (D2) in integument and haemolymph, respectively. On the other hand, zinc showed high content at premolt stage in haemolymph and early postmolt stage in gill supporting its function in calcification. The results confirmed the roles of chitobiase, proteinase, glycogen, and trace elements which corresponded well with molting stages and physiological changes of mud crab.
