หัวเรื่อง:ไม่มีชื่อไทย (ชื่ออังกฤษ : Morphology and Cytochemistry of Blood Cells from Asian Wild Dog (Cuon alpinus)) ผู้เขียน:ดร.เฉลียว ศาลากิจ, ศาสตราจารย์, นายเจริญศักดิ์ ศาลากิจ, รองศาสตราจารย์, นางสาวจุฑามาส รัตนคุณูประการ, Narit Tengchaisri, Wanchai Tunwattana, สุนทรี อภิบาล สื่อสิ่งพิมพ์:pdf AbstractBlood cells from direct blood smears of four adult Asian wild dogs (Cuon alpinus) were examined and measured after stainning with modified Wrights Giemsa stain and cytochemical stains, including Sudan black B (SBB), ?-naphthyl acetate esterase (ANAE), and ?-glucuronidase (?-glu). Red blood cells show uniform in sized, biconcave and prominent central pallor with 6.0 ?m mean diameter. Reticulocytes and nucleated red blood cells stained positive for ANAE and ?-glu. Neutrophils have tight constricted and multilobulated nucleus which mostly occupied the cells. Neutrophils stain strongly positive with SBB, negative for ANAE and weak positive for ?-glu. Eosinophils contain numerous round red refractile granules with some vacuoles. Eosinophils stain moderately positive for SBB and ?-glu but negative for ANAE. Basophils have variable numbers of intensely basophilic granules which do not obscure the very long lobulated nucleus. Basophils stain moderately positive for SBB but strongly positive for ANAE and ?-glu. Most lymphocytes are small and medium size (65:68). Lymphocytes are negative for SBB but have 3 patterns of reactivity for ANAE and ?-glu, including negative, focal dot staining and fine granular staining. Monocytes stain moderately positive for SBB and moderately to strongly positive for ANAE and ?-glu. Platelets are negative for SBB and stain moderately to strongly positive for ANAE and ?-glu. |
หัวเรื่อง:ไม่มีชื่อไทย (ชื่ออังกฤษ : Hematology, Cytochemistry and Ultrastructure of Blood Cells from Asian Elephant (Elephas maximus)) ผู้เขียน:นายเจริญศักดิ์ ศาลากิจ, รองศาสตราจารย์, ดร.เฉลียว ศาลากิจ, ศาสตราจารย์, นางสาวนวลอนงค์ นาคคง, สุนทรี อภิบาล, Piyawan Suthunmapinuntra, Jutamat Rattanakukuprakarn, Gavil Nunklang, Manoch Yindee สื่อสิ่งพิมพ์:pdf AbstractBlood cells from fourteen (5 males and 9 females) adult Asian elephants were examined and cytochemical stained with Sudan Black B (SBB), peroxidase, periodic acid Schiff’s reaction (PAS), anaphthyl acetate esterase (ANAE) and b-glucuronidase (b-glu). The complete blood counts were performed using automate cell counter. There were no significant differences nearly all hematological values between the male and female elephants except the leukocyte count and fibrinogen concentration which were higher and lower, respectively, in the males than the females. Neutrophils had poorly segmented neclei and many well-differentiated granules. Neutrophils stained strongly positive to SBB, faintly stained with PAS, focal dot stained to ANAE and b-glu. Eosinophils contained 2-3 lobe nuclei, numerous small round red refractive granules with some vacuoles. Eosinophils stained moderately positive to SBB and strongly positive to ANAE but negative to b-glu. Basophils had variable number of intensely basophilic granules which did not obscure the lobed nuclei. Basophils were negative for SBB but moderately positive to ANAE and b-glu. Monocytes stained moderately positive to SBB and moderately to strongly positive to ANAE and b-glu. The bilobed cells stained moderately positive for SBB and strongly positive for ANAE and b-glu which were similar to monocytes. Ultrastructurally, they contained a large number of mitochondria similar to those of monocytes except the shape of the nuclei. The number of bilobed cells exceeded the number of the other leukocytes. Scanning electron microscopy revealed surfaces of all blood cells. Transmission electron microscopy revealed organelles within erythrocytes, platelets and all leukocytes especially bilobed cells. |

There was no blood parasite in all civets. The red blood cells showed relatively uniform in size, with 4.3 ?m mean diameter. Neutrophils had tight constricted, multilobed nuclei and contained many specific granules which clearly detected by transmission electron microscope (TEM). Neutrophils stained strongly positive with Sudan black B (SBB) and peroxidase (PO) but negative with alpha-naphthyl acetate esterase (ANAE) and ?-glucuronidase (?G). By scanning electron microscope (SEM), they showed many microvilli and some micropores. Eosinophils contained numerous large round red refractive granules and stained strongly positive with SBB and PO, but negative with ANAE and ?G. By SEM, the granule contour of eosinophil was most easily delineated from the other granulocytes. Ultrastructurally, they contained large round granules with homogenous content without crystalloid-core. Basophils had variable numbers of dull grey, pleomorphic granules and were negative for SBB and PO, but strongly positive for ANAE and moderately positive for ?G. By SEM, the surface of basophil was smooth with small granule contour. Ultrastructurally, they contained smaller, homogenous and less electron-dense granules than those in the eosinophils. Lymphocytes were negative for SBB and PO but had 2 patterns of reactivity for ANAE and ?G, including focal dot and fine granular staining. Monocytes were negative for SBB, PO and ?G but moderately positive for ANAE. The large round eosinophil granules were the characteristic of leukocytes in common palm civet. |

Red blood cells were uniform in shape, with 7.3 mm mean diameter in size and easy to form rouleaux. Using scanning electron microscopic (SEM) examinations revealed normal and abnormal red blood cell surfaces. Neutrophils contained several vacuoles as detected by light microscope and revealed themselves as large granules under transmission electron microscope. Neutrophils stained strongly positive with SBB, ANAE; weak positive with PAS and negative with AcP and b-glu. Using SEM, neutrophil surfaces revealed several microvilli and some micropores. Eosinophils contained numerous small round red refractive granules with some vacuoles. Eosinophils stained strongly positive with SBB and ANAE but negative with PAS and b-glu. Basophils had variable numbers of intense basophilic granules that obscured the very long lobulated nucleus. Basophils stained moderately positive with SBB but strongly positive with ANAE. Lymphocytes were negative with SBB but have 3 patterns of reactivity with ANAE, AcP and b-glu, including negative, focal dot and fine granular stainings. Monocytes stained moderately with SBB and moderately to strongly with for ANAE and b-glu. The SEM examinations could differentiate white blood cells by their surface contours. Transmission electron microscopic examinations revealed organelles within all blood cells. |
หัวเรื่อง:ไม่มีชื่อไทย (ชื่ออังกฤษ : Hematology, Morphology, Cytochemistry and Ultrastructure of Blood Cells in Painted Stork (Mycteria leucocephala)) ผู้เขียน:ดร.เฉลียว ศาลากิจ, ศาสตราจารย์, นายเจริญศักดิ์ ศาลากิจ, รองศาสตราจารย์, นางสาวนวลอนงค์ นาคคง, Decha Pitakkingthong, ทรงกลด ภู่ทอง สื่อสิ่งพิมพ์:pdf AbstractBlood samples of 12 (7 males and 5 females) painted storks (Mycteria leucocephala), at the Bangpra Waterbird Breeding and Research Center, were collected from the cutaneus ulna vein for basic hematologic, light microscopic and transmission electron microscopic studies of blood cells. There was no hematozoa detected in all painted storks. There was no significant difference in all hematological values between the male and female groups. Lymphocytes were the most commonly observed leukocytes in the male painted storks and average 6-8 mm in diameter. Heterophils were the most commonly observed leukocytes in the females and were the second most commonly observed leukocytes in the males. Heterophils contained many pleomorphic, dull eosinophilic granules, ribosomes and lobed nucleus. Heterophils were the medium-sized cells, average 8-12 mm in diameter. Heterophils stained strongly positive with a-naphthyl acetate esterase (ANAE). Eosinophils revealed many small round granules and usually were the largest leukocytes, average 9-14 mm in diameter. The number of eosinophils was quite high in both the male and female painted storks. Eosinophil granules stained intensely with b– glucuronidase (b–glu), moderately to strong stained with ANAE and also stained with new methylene blue. Basophils were as small as lymphocytes but revealed many small granules surrounded central round nucleus. Basophils stained moderately with b–glu. Transmission electron microscopic examination revealed organelles within erythrocytes, heterophils, eosinophils and basophils |
