Semen of 20 Holstein-Fresian bulls aged 3-5 years were collected and frozen once a week for 3 weeks. The result showed the post-thaw sperm motility and progressive motility at 0 h from the 87% and 99% glycerol were not significantly different (P>0.05). The ALH (amplitude of lateral head displacement) and BCF (beat cross frequency) of the sperm from the extender using the 99% glycerol was significantly higher (P<0.05). However, after 5 h of incubation at 37?C the 87% glycerol gave significantly lower motility and progressive motiloty with higher VAP (average path velocity), VCL (curvilinear velocity) and ALH (P<0.05). The results of dilution methods did not differ significantly at 0 h. except at 5 h. the one-step method revealed significant higher motility (P<0.05. There were no effects of glycerol cencentration and dilution methods on post-thaw sperm head elongation, area, plasma membrane and acrosomal integrity (P>0.05). In conclusion, using 99% glycerol and one-step dilution methods gave a better semen quality. |

Plantlets were transferred to incubator controlled at 5?C and 15?C under 24 hours light condition for I and 2 months. After the storage periods, plantlets were cultured under room temperature for I month. Culture under 5?C for 1 month and 15?C for 1 and 2 months could decrease growth of sugarcane plantlets in which all of plantlets could normally rcgrowth under room temperature. Plantlets of sugarcane cultivar Supan I cultured under 5?C for 2 months could. regrowth only 50-60% under room temperature. |
