Tissue explants from root tip and mesocotyl were suitable for callus formation. The induction and growth of callus was better in medium containing 1 mg/1 2, 4-D and 10% coconut milk than medium containing 3 or mg/1 2, 4-D and coconut milk. Callus from root tip which was cultured in medium containing 1 or mg/1 2, 4-D grew at a greater extent under condition of light than in the dark. In contrast, callus from mesocotyl which was cultured in medium containing 1 mg/1 2, 4-D grew better under condition of dark than in the light. Nevertheless, callus grew better in the dark in medium which was supplemented with %coconut milk. The increasing percentage of callus growth was greater in medium containing 3 mg/1 2,4-D and 10% coconut milk or in coconut milk free-medium. Root formation occurred in callus which was cultured in medium containing 1 or 3 mg/1 2,4-D than in medium containing 1 mg/1 2,4-D than in medium containing 3 mg/1 2,4-D. |

The medium which consisted of MS macro and micro-elements, organic addenda and 0.05 ppm naphthaleneacetic acid (NAA) was supplemented with 6-benzylamino purine (BA) at 0.1, 0.2, 0.3, 0.4, 0.5 ppm (Experiment 1) or 6-furfurylamino purine (kinetin) 0.5, 1.0, 1.5, 2.0, or 2.5 ppm (Experiment 2). BA 0.1 ppm (Experiment 1) promoted the longest shoot and root and the highest number of leaves and roots, but tended to reduce number of branch. BA 0.5 ppm caused lesser number of branch and rroot to form than BA 0.1 ppm, while BA 0.4 ppm caused the least root formation. BA 0.3 ppm however promoted the most number of shoot branching. Kinetin 0.5 ppm (Experiment 2) promoted the longest shoot and the highest number of leaves and survival explants, but caused the least branching. Meanwhile, kinetin 2 ppm promoted the most number of branching. |

They were able to regenerate plantlets. Shoot-tip tissues of A. amazonica were able to regenerate 1,295 plantlets per shoot-tip within 6 months while A. chelsonii regenerale 1,369 plantlets per shoot- tip. However. A. amazonica could produce callus 1 2.05% of the cultured tissues after 3 months, of which it could be induced to regenerate plantlets as well. Plantlets from these 2 species could be obtained within 30-40 days after culturing in vitro |

Five varieties, Caroussel, Excelsior, Scarlet Leader, Rembrandt and Rilona, were able to regenerate bulblets. Excelsior tissues formed callus 90% and regenerate bulblet 50%. Both Rilona and Caroussel tissues formed callus 70% and bulblet 30%. Regeneration of bulblet in four varieties took 60-70 days but Caroussel variety took 32 days. |
