Abstract

The efficiency of the selection procedure during succeeding generations is the most important role of any breeding program. There are many selection methods, but no method is perfect for general use with all crop plants. Breeders must carefully consider which method will be the most effective for their purposes. This study was conducted to compare the effectiveness between the pedigree selection (PS) and single seed descent (SSD) methods in two yardlong bean crosses. Both procedures started from the same F2 population in each cross. In 2004, 15 F4 progenies were selected by each selection method from each population. Thirty F4 progenies and the parents of each population were tested in separate experiments with two check cultivars at the Songkhla Field Crop Research Station, Songkhla Province. Selected-PSU and VU135 were grown as check yardlong bean cultivars. Narrow sense heritability (h2) for the pod yield and yield components were estimated through regression analysis of the F4 progenies on F3 parental plants. Correlations among yield and yield components were also estimated. The results showed that the mean pod yield and yield components of selected F4 progenies derived from both PS and SSD were not significantly different in both the 4501 and 4502 populations. Low heritability for pod yield per plant was found in both the 4501 and 4502 populations at 4.62 and 2.96%, respectively, which indicated why selection for yield improvement in early generations had been ineffective. The highest positive correlation was found between the number of pods per plant and pod yield in both the 4501 and 4502 populations, with correlation coefficients of 0.7540 and 0.9229 (p<0.01), respectively. This study shows that PS and SSD are equally effective for yield improvement in yardlong bean.

Abstract

Cultivation problems of neck oranges decline at Amphur Chana, Songkhla Province, Were investigated. As essential nutrient elements were the first suspected problem, bulk soil samples around the problem site were randomly collected. The soil was mixed, sieved and employed for all glasshouse experiments. There were to find a suitable rate of basal nutrient, to evaluate fertility status of the soil and suitable rates of individual nutrient ele?ments. Maize was used as the tested plant and its shoot dry weight was employed as a measured parameter. Results demonstrated 3xAll of the basal nutrient element gave best growth of maize grown on this soil type. The soil was deficient in N, P, S, Cu, Mo and Zn. Hence, rates of these nutrient elements were studied. It was found that maximum corresponding rates for N, P, S, Cu, Mo and Zn were 120, 150, 75, 6, 0.8 and 5 kg ele?ment/ha respectively. There after, these rates of elements were used in a farmer plot to test whether these ele?ments would help in preventing the decline in growth of the neck orange.

Abstract

Rubber seedlings from two early-introduced clones (EIRpsu 1 and EIRpsu 2) in southern Thailand were evaluated for root growth performance under field conditions. The two clones had been screened for white root disease tolerance. Genetic analysis of EIRpsu 1 and EIRpsu 2 was compared to RRIM 600 (the major cultivated variety of Hevea rubber in Thailand) using random amplified polymorphic DNA markers with 10 primers. Results from cluster analysis indicated that EIRpsu 2 was closer to RRIM 600 than EIRpsu 1 with similarity coefficients of 0.850, 0.860 and 0.890, respectively. Root growth of EIRpsu1 and EIRpsu2 was monitored at a rubber plantation in Langsuan district, Chumphon province during 2012–2013. Six-month-old seedlings of each clone were transplanted into a rhizobox. The experiment was designed as a randomized complete block design with four replications, with one plant per rhizobox. Shoot growth and root proliferation of the seedlings were recorded at two-monthly intervals from October 2012 to March 2013. The shoot growth was monitored by measuring plant height, trunk diameter and the number of compound leaves. It was found that RRIM 600 exhibited the highest shoot growth under field conditions, and it was significantly different from the other two clones. The root length density was measured by scanning the root systems of each seedling from a panel in the rhizobox. Results showed that the seedlings of EIRpsu 1 and EIRpsu 2 had better root growth than RRIM 600. The spatial distribution of roots indicated a rather deeper root system for the two selected clones.

Abstract

The coat protein gene of Blackeye cowpea mosaic virus (BICMV-CP) was amplifi ed by reverse transcription polymerase chain reaction and cloned into the expression vector pQE-80L. This plasmid was transformed into Escherichia coli DH5? competent cells. The BICMV-CP gene was expressed as a fusion protein containing a fragment of 6xHis-tag. Bacterial cells were disrupted by repeated freezethawing three times and the BICMV-CP fusion proteins were purifi ed under denaturing conditions by affi nity chromatography with Ni-NTA Agarose. A sample of 500 ?g of purifi ed protein was mixed with Freund’s complete adjuvant at a ratio of 1:1 (volume to volume). Initially, the emulsion was subcutaneously injected into a New Zealand White rabbit, followed at weekly intervals by three additional immunizations with 500 ?g of the purifi ed protein mixed with Freund’s incomplete adjuvant. Bleeding was done every week during weeks 5-12 and titers of the antisera ranging from 800-51,200 were obtained. Up to a dilution of 1:320 of the BICMV-infected yardlong bean sap could be detected by indirect enzyme-linked immunosorbent assay. The produced antiserum reacted specifi cally with the BICMV-infected plant without any cross reaction with other virus species tested. However, a weakly positive reaction with Bean common mosaic virus could be observed.
