Abstract

A total of forty-six lactic acid bacteria (LAB) strains were screened for production of antibacterial substances, out of which two strains, JAK-CS7-1 and JAK-M1-3, displayed broad inhibition spectra against Gram-positive and Gram-negative pathogenic bacteria. Based on physiological, biochemical and molecular methods, these two bioactive thermophilic LAB strains were identified as Pediococcus acidilactici. The amounts of lactic and acetic acids produced by these two LAB strains were determined by HPLC method and then used for preparing the organic acid model system performance (OMP). Thus, comparative evaluation of the antibacterial activities of the culture supernatants (CS) and the corresponding OMP, consisting of equivalent acids concentration and similar pH like CS, revealed that the OMP displayed lower activities than the CS. It was found that the antibacterial activities of the CS were greater than the OMP by 50% that could be attributed to the presence of additional antibacterial metabolites in the CS. The results of this study suggested that these two strains of Pediococcus acidilactici appear to contain other bioactive substances in addition to lactic and acetic acids.

Abstract

Blood samples of 12 (7 males and 5 females) painted storks (Mycteria leucocephala), at the Bangpra Waterbird Breeding and Research Center, were collected from the cutaneus ulna vein for basic hematologic, light microscopic and transmission electron microscopic studies of blood cells. There was no hematozoa detected in all painted storks. There was no significant difference in all hematological values between the male and female groups. Lymphocytes were the most commonly observed leukocytes in the male painted storks and average 6-8 mm in diameter. Heterophils were the most commonly observed leukocytes in the females and were the second most commonly observed leukocytes in the males. Heterophils contained many pleomorphic, dull eosinophilic granules, ribosomes and lobed nucleus. Heterophils were the medium-sized cells, average 8-12 mm in diameter. Heterophils stained strongly positive with a-naphthyl acetate esterase (ANAE). Eosinophils revealed many small round granules and usually were the largest leukocytes, average 9-14 mm in diameter. The number of eosinophils was quite high in both the male and female painted storks. Eosinophil granules stained intensely with b– glucuronidase (b–glu), moderately to strong stained with ANAE and also stained with new methylene blue. Basophils were as small as lymphocytes but revealed many small granules surrounded central round nucleus. Basophils stained moderately with b–glu. Transmission electron microscopic examination revealed organelles within erythrocytes, heterophils, eosinophils and basophils

Abstract

This paper presents a packing algorithm to solve an irregular shape strip packing problem. The polygons are packed in strip material, which limits a bin width but not limits a bin length. The objective of a packing algorithm is to minimize a bin length. This paper proposes a hybrid method of genetic algorithms and tabu search to solve a packing problem. Initially, a hybrid method behaves like regular genetic algorithms. After that, a hybrid method is self adapting until it behaves like a tabu search. This paper compares results generated by a hybrid method with genetic algorithms and tabu search. The experiment data is the 13 pieces convex-type polygon and duplicate, triplicate and quadruplicate. The experiment shows that a hybrid method generates better result than genetic algorithms about 2.56% to 4.69% and a hybrid method generates better result than tabu search about 0.26% to 2.78%.

Abstract

This research aimed to develop the multireservoir water allocation model for the Upper Mun basin in Northeast of Thailand where there were 4 large scale multipurpose reservoirs namely Lam Chae, Mun Bon, Lam Phra Ploeng and Lam Takong located. The study area covered 7,190 sq.km. The water in the basin was used for 4 purposes namely agriculture, municipal and industrial water supply, preserving the ecological system equilibrium and the water requirements at the river basin outlet for downstream users. There were 4 big irrigation systems covering the area of 353,650 rai and 8 municipal and industrial water supply systems. 25 years of monthly data were used in the analysis of the water shortage in the basin. The optimum water allocation among the 4 water user groups was derived by the multiobjective optimization techniques called Epsilon-constraint linear programming. The best water allocation policy that satisfied the profitability, equity and reliability criteria was derived by the multicriteria decision making process called AHP. The priority weights for profitability, reliability and equity were 41, 32.3 and 26.7% respectively. The best water allocation policy was then simulated by HEC-3 using the cases of dry and normal year. The simulation result was used to develop the artificial neural networks model for multireservoir water allocation. The 12-16-4-4 MLFF-BP neural networks were the best fitted model. The model showed very high R2 of 0.9578-0.9900 for training and R2 of 0.8041-0.9658 for the testing. The 12-16-4-4 MLFF-BP neural networks model was tested by comparison with the actual data in 2001 and 2002. This research illustrated that the artificial neural networks was simple and could practically be applied to the multireservoir water allocation.

Abstract

Entrapment in sol-gel from rice husk ash was established by immobilization of alkali lipase produced by Pseudomonas sp. KLB1 lipase. The enzyme was strongly immobilized by entrapment in neutral sol- gel of rice husk ash. It was optimally active at 60?C and pH 9. Its activity was constant in the medium pH 9 under temperature 70?C for 1 hr. The immobilized lipase was activated by Ca2+, K+, Na+, EDTA and ascorbic acid but inhibited by Fe2+, Mn2+, Co2+, and KI. Both Ca2+,and Na+ were mainly found in rice husk ash and xerogel. The effective concentration of CaCl2 and NaCl on the enzyme activation were 60 mM and 20mM, respectively. According to reusability, it was applied into the hydrolysis of p-nitrophenyl palmitate by 12 times which lost 50% activity. The Km, and Vmax of p-NPP hydrolysis catalyzed by the entrapped enzyme were 7.43 ? 10–2 mM, and 1.94 ? 10-5 mM s-1, respectively, whilst by the free enzyme were 21.69 mM, and 3.90 ? 10-3 mM s-1, respectively.

Abstract

The Effectiveness of EM for controlling asian citrus psyllid. Diaphorina citri Kuwayama was conducted at Entomology and Zoology Division, Department of Agriculture during August to December 1994. The EM was tested for its insecticidal and repellent properties on the 2nd and 3rd nymphal stages. The experiment consisted of 4 treatments : including 1.) spraying with mixed solution of molasses, ethyl alcohol, acetic acid, EM and Water (ratio 1:1:1:1:10, at 15 days fermentation) 2.) spraying with the mixed solution as same as above but without EM 3.) spraying with water only and 4.) untreated. The result showed that after spraying 3 days, psyllid nymphs in all treatments were not be killed and they also develops into the next growing stage. Moreover, EM had on property on repellent to the asian citrus psyllid. It was concluded that EM was not effective in controlling asian citrus psyllid.

Abstract

Insecticidal and antifeedant test of EM to the leaf roller, (Lamprosema diemenatis (Guenee)) were studied and carried out in the laboratory during April 1994. Insecticidal study of EM on the third instar larvae showed non significantly different between the toxicity of solution A (mixed solution with EM5) and solution B (mixed solution with out EM5) or control (sprayed with fresh water or on spraying). In the case of antifeedant test it found that, the larvae could less damage on the leaf sprayed with the two mixed solutions. But the percentage of leaf damage was not significantly different between the leaf sprayed with solution A and B.

Abstract

The effect of EM 5 on African red mite, Eutetranychus africanus (Tucker) Citrus rust mite, Phyllocoptruta oleivora (Ashamead) and Citrus yellow mite, Eotetranychus cendanai (Rimando) were examined in the laboratory of Entomology and Zoology Division. This study was made to determine the possibility of developing some effective microorganisms to control the important citrus mites. The EM 5 mixture including30% ethyl alcohol, acetic acid and molasses were diluted with water into 5 concentrations : 100, 75. 50, 25 and 12.5%. The pure mixture excluded EM and a recommended acaricide, propargite 0.045% were included to test the toxicity on these as well. All treatments were evaluated by use of the leaf disc-dip technique. The residual actions on mite mortality were determined 24, 48, 72 and 96 hours after treated. Excluded the errors of this experiment, the result would conducted on 48 hours after treated, The mite mortality treated with propargite 0.045% was one hundred percentage which was non significantly different from mixture plus EM 100% and mixture without EM 100%. Because the 100% mixture with and without EM gave the some effect on mite mortality, the result was concluded that the toxicity to mite was not caused by EM. They might be killed due to the compounds in mixture, such as acetic acid, ethyl alcohol or due to the very high sticky of molasses.
