Changes in the chemical composition of the intestinal mucous affect the functioning of the large intestine. The expression of glycoconjugates in the colonic epithelium of buffaloes was studied using histochemical methods. Paraffin sections 3 ?m thick were stained with the following reagents: 1) haematoxylin and eosin (H&E), 2) alcian blue (AB) pH 2.5, 3) periodic acid –Schiff (PAS), 4) AB pH 2.5 – PAS, 5) high iron diamine (HID), 6) HID–AB pH 2.5, and three lectins, 7) Arachis hypogeae (PNA), 8) Ulex europaeus (UEA-I) and 9) Triticum vulgaris (WGA). Neuraminidase was used before staining with PNA. The histology of the descending colonic epithelium is characterized by an absence of villi; the epithelium have numerous simple columnar crypts. The crypts consist of columnar absorptive cells and goblet cells. The goblet cells in the descending colon of buffaloes contained acid and neutral glycoconjugates, whose expression was increased from the lower crypts to the upper crypts. There were more sulphated glycoconjugates in the goblet cells of lower crypts than in upper crypts. The crypts of the upper areas showed more intense expression of carboxylated glycoconjugates and sialic acid together with D- galactose and N- acetylglucosamine. The expression of ? -L- fucose was found in the goblet cells throughout all crypts. The results indicated that sulfomucin and ? -L- fucose played a lubricating role, while sialic acid and sialoderivatives provided protection against pathogenic infection. |

The staining procedures employed were horseradish peroxidase conjugated lectin, alcian blue (AB) pH 2.5, periodic acid-Schiff (PAS), ABpH 2.5-PAS, high iron diamine (HID), HID-ABpH 2.5. The lectins used in the present study were concanavalin A (ConA), Ricinus communis agglutinin I(RCA-I), Solanum tuberosum agglutinin (STL) and Limulus polyphemus agglutinin(LPA) . The result revealed that descending colonic epithelium of the goat contained acid sulfated glycoconjugates and neutral glycoconjugates with ?-D-mannose, ?-D-glucose, ?-D-galactose, Nacetylglucosamine and sialic acid residues. The quantity of neutral glycoconjugates with ?-D-mannose, ?-D-glucose, ?-D-galactose and N-acetylglucosamine increased from mucosal crypt up to mucosal surface of the epithelium, while sialic acid residues and sulfated glycoconjugates decreased from mucosal crypt to mucosal surface. |

Observation of cross sections by light microscope showed the presence of vascular occlusion in aged flower stems. Occlusions at the basal portion were much more than at the upper portion. Occluding material at both the basal and upper portions above the water level contained both pectin and carbohydrate, while only at the upper portion contained phenol, Lignins were not found in the occluding material. |

The staining procedures employed were alcian blue (AB) pH 1.0 AB pH 2.5, Periodic acid Schiff (PAS) and AB pH 2.5-PAS. In addition twelve lectins, Triticum vulgaris WGA, Concana valin A, Glycine max SBA, Dolichos biflorus DBA, Arachis hypogea PNA, Solanum tuberosum STL, Datura stramonium DSA, Ulex europeus UEA-I, Bauhinia purpurea BPA, Ricinus communis RCAI, Lotus tetragonolobus LTA, and Limulus polyphemus LPA were applied to detect saccharides residues. According to the results obtained, the nature of glycoconjugates in mucous epithelium was found to change during cell migration from deep to superficial part. The surface mucous cells contained primarily neutral glycoconjugates with vicinal diol groupings and terminal fucose residues. By contrast, the mucous cells in deep pits and in pyloric glands contained acidic sulfated glycoconjugates with terminal sialic acid residues. The ?-D-mannose, ?-D-glucose, ?-D-galactose, N-acetylglucosamine and N-acetylgalactosamine were also found at the pyloric gland cells while they were smaller in amount at the surface mucous cells. The data suggested that changes of epithelial cells were closely correlated with cellular maturation. |

A series of staining procedures were employed including peroxidase - labeled lectin - diaminobenzidine methods and digestion techniques with neuraminidase or ? - amylase. The terminal portion of the anterior lingual gland consisted of mucous and seromucous cells. All the mucous cells in the glands possessed a mixture of glycoconjugates of different nature, but they could be divided into three types, on the basis of the principle type of glycoconjugates involved, namely primarily sulfated, carboxylated and neutral glycoconjugate-containing cells. The first mentioned type of cells was predominant. The presence of sialic acid, ? - D - mannose, ? - D - glucose and ? - D - galactose in these glycoconjugates was confirmed. The seromucous cells found in the anterior lingual gland contained a very small amount of comparable carbohydrates predominantly in the apical cytoplasm. Possible functional activities of the glands were discussed. |
