The effects of trehalose supplementation (with and without trehalose supplementation; 100 mmol/l), equilibration times (30 min or 60 min) and boars differences on in vitro post-thaw sperm quality were determined. Fresh semen was collected from 4 Duroc boars (age 2.5 1 years) (Fresh semen was collected 4,3,3,2 times in each boar) diluted with the Beltsville Thawing Solution extender and kipt at 15?C for 2 h. After centrifugation, the supernatant was discarded and diluted the sperm pellet with the cooling media by adjusting the concentration to 1.5 x 10 sperm/ml, then held at 5 ?C for further 1.5 h. Diluted semen was divided into 2 groups depended on the types of freezing media. Each group was equilibrated by incubation at 5?C before loading into 0.5 ml straw. The straws were placed on liquid nitrogen vapor for 20 min and then plunged into liquid nitrogen. Frozen semen was thawed by immersing in 37?C water for 30 sec and evaluated sperm quality. The results revealed that trehalose supplementation and equilibration time had no effects on post-thaw sperm quality (p>0.05). Only boar differences affected progressive motility (p = 0.043),average path velocity (p=0.003) and straight-line velocity (p =0.032). This study demonstrated that boar differences was the main factor influenced frozen- thawed semen quality.
