Abstract

A transformation experiment was carried out by optimizing three important parameters: the age of calli, infection time and the optimum density of Agrobacterium suspension. A high efficiency Agrobacterium-mediated transformation system was established for the indica rice Hom Kra Dang Ngah variety. Embryogenic calli with high regeneration capability were incubated under different conditions with Agrobacterium tumefaciens strain EHA105 harboring plasmid pCAMBIA1304-EPSPs. The plasmid contains the enzyme gene 5-Enolpyruvylshikimate 3-phosphate synthase (epsps) as a selection marker gene and ?-glucuronidase (gus) as a reporter gene. After 2 wk of Agrobacterium elimination and 4 wk of culturing on a selection medium containing 0.5 mM glyphosate (subcultured at 2-weekly intervals) the results showed that the six-week-old calli infected with bacterial suspension at OD600 0.6 for 20 min gave the highest frequency of gus gene expression of 83.5%. The target gene was confirmed using polymerase chain reaction.

Abstract

Hevea brasiliensis is an economically important perennial tree in southeast Asia. However, few studies have been conducted to evaluate the effects of sucrose and genotype on callus induction and plantlet regeneration from anther culture. Male flowers at the uninucleate stage were collected from clones numbered 2-nr, 6-nr, 1-tF and 1-em. An excised cluster of anthers was cultured on callus induction medium (CIM) consisting of Murashige and Skoog (MS) medium supplemented with 1 mg.L-1 2,4-diclorophenoxyacetic acid, 1 mg.L-1 kinetin (KN), 1 mg.L-1 ?-naphthalene acetic acid (NAA) and sucrose at various concentrations ranging from 3 to 10%. The results revealed that the highest callus induction frequency (87.5%) was obtained from clone 1-em on CIM with 8% sucrose. Callus derived from clone 2-nr culturing on CIM supplemented with 5% sucrose gave the highest frequency and number of somatic embryo (SEm) formations. CIM supplemented with 5% sucrose and 5% coconut water caused a decrease in the percentage of callus induction (30.88%). The highest SEm formation at 20% was obtained when callus of clone 2-nr aged 8 wk was cultured on MS supplemented with 3% sucrose, 0.2 mg.L-1 NAA, 1 mg.L-1 6-benzyladenine, 3 mg.L-1 KN and 0.05 mg.L-1 gibberellic acid (GA3). When calluses were cultured on CIM supplemented with 5% sucrose and maintained in darkness, 16% SEm formation was obtained. Calluses and SEms from the culture medium showed the same ploidy level as the mother plant after evaluation by flow cytometry.

Abstract

The objective of this study was to determine the impact of different rain protectors and varieties of broccoli on their growth and yield during the rainy season in southern Thailand. Split plots in a randomized complete block design were used with four replications. The experiment was undertaken from 28 October 2012 to 7 January 2013. The results showed that broccoli grown under a green shade net or a plastic sheet had higher growth and yield compared with those grown in the open field. The average highest total yields under plastic sheet, green shade net and in the open field were 11.11, 7.54 and 5.61 t.ha-1, respectively. The average highest yields of broccoli were obtained from the Yok Kheo variety (10.22 t.ha-1), followed by the Green Queen (7.56 t.ha-1) and Top Green (6.46 t.ha-1) varieties, respectively. Broccoli grown under the plastic sheet could be harvested 13.25 d after transplanting and 2.50 d earlier than those grown in the open field or under green shade net. The results indicated that broccoli should be grown under a plastic sheet or a green shade net to achieve higher yields than from plants grown in the open field. The Yok Kheo and Green Queen varieties are recommended for Songkhla province and the surrounding area.

Abstract

The infl uence was investigated of the hygromycin concentration on the survival rate of oil palm secondary somatic embryos (SSEs) and sonication-assisted Agrobacterium-mediated transformation (SAAT). The results showed that 20 mg.L-1 hygromycin could completely inhibit growth of SSEs, suitable for selection of transformed SSEs. The transient ?-glucuronidase (gus) expression was affected by the different period of SAAT which was signifi cantly enhanced by increasing the period of SAAT. However, the longer exposure to SAAT caused a decrement in the frequency of plantlet regeneration. The highest transformation effi ciency was obtained when the SSEs were sonicated for 5 min. Polymerase chain reaction and Southern blot hybridization analysis confi rmed the presence of the reporter genes in the transformed plantlet genome.
