It was found that the MS me?dium supplemented with 0.2 mg/1 BA and 0.02 mg/1 NAA gave the best proliferation rate, 4-5 shootlets could be obtained within 20 days from development of axillary buds. After 50 days the shootlets were multiplied at the rate of 4 folds in every 2 weeks by subculturing into the same medium The resulting shootlets could be progres?sively increased at the rate of 6 folds in every 4 weeks when transferred on to an MS solid medium, supple?mented with 0.2 mg/l BA and 0.02 mg/1 NAA before transferred on to MS medium containing 15oh (v/v) coco?nut water and 1 mg/ l 2,4-D to enhance the shoots. Rooting of the shoots were successfully done on a MS me?dium with 1 mg/I IBA. |

This callus was preserved under low temperature at 1 and 5 degree C with different periods of preservation, and then was cultured at 27 degree C for 6 months. After preservation under low temperature, plantlets could be induced from callus preserved at 5 degree C but could not be induced from callus preserved at 0 degree C. In addition, at 5 degree C callus could develop to be plantlets only when preserved not longer than 30 days. Callus preserved at low temperature grew slower than callus not preserved at low temperature. |

Line numbers 2156, 58608, 2160 and 2180 expressed short stature and early maturity. Their heights at maturity time were shorter that the original Doi Kham for 13, 15, 17 and 22 cm respectively, and their yields were 13, 11, 11 and 13 days earlier than Doi Kham, respectively. Their yields were similar to Doi Kham. These four lines will be further tested at different locations in the dry and rainy seasons. The best line will be selected and used as a variety. |
