Fungfupong, P. Srinives สื่อสิ่งพิมพ์:pdf AbstractThe anthers of F1 hybrid rice (Khao Dawk Mali 105/Skybonnet) were cultured on the various callus induction media. It was found that a suitable medium for callus induction was N6 containing 2 mg/l 2,4-D, 1 mg/l kinetin, 500 mg/l casein hydrolysate and 50 g/l sucrose. The plantlets could be produced from callus on the MS medium supplemented with 2 mg/l BAP, 1 mg/l kinetin, 0.2 mg/l NAA, 300 mg/l casein hydrolysate, 15% coconut water and 20 g/l sucrose. After transferring these plantlets onto the MS medium containing 1 g/l yeast extract, 15% coconut water, 3 mg/l BAP and 0.01% colchicines for 5 days, about 60% of the doubled haploid plants were recovered. |

Klakhaeng สื่อสิ่งพิมพ์:pdf AbstractThe panicles of the F1 hybrid rice (Lemont/Khao Dawk mali 105) of which the distance between the auricles of the flag leaf and the subtending leaf was 6-12 cm were selected for anther culture. These spikelets had anthers bearing the mid-uninucleate microspores. High percentage of callus formation was obtained when anthers were cultured on the N6 medium supplemented with 500 mg/l casein hydrolysate, 2 mg/l NAA, 1 mg/1 kinetin and 50 g/l maltose. The calli could regenerate into plantlets at high frequency when cultured on the MS medium containing 15% coconut water, 300 mg/l casein hydrolysate, 20 g/l sucrose, 2 mg/lBAP and 0.2 mg/l NAA. About forty percent and forty-two percent of double haploid plants were obtained when the regenerants were transferred on the MS medium containing 1 g/l yeast extract, 15% coconut water, 3 mg/ 1 BAP and 0.01% colchicines for 3 and 5 days, respectively. |

The suitable callus induction medium was the modified MS medium containign I mg/l 2,4-D, 2 mg/l NAA and 1 mg/l kinetin. Calli from this medium were divided into two groups, one was 3 week old calli (non subcultured calli) and the other was 6 week old calli (subcultured calli). Both groups seemed to grow and develop into plantles quite well on the modified MS medium supplemented with 2 mg/l kinetin. However the percentage of plantles induction from 3 week old calli was better. Calli of both groups were irradiated with different doses of gamma radiation and were cultured on the regenerated medium. The 3 week old calli were better than 6 week old calli in regenerating into plantles. |
